Natural variations in a pectin acetylesterase gene, <i>MdPAE10</i> , contribute to prolonged apple fruit shelf life

چکیده
Room-temperature shelf life is a key factor in fresh market apple (Malus domestica Borkh.) quality and commercial value. To investigate the genetic molecular mechanism underlying life, quantitative trait loci (QTL) were identified using bulked segregant analysis via sequencing (BSA-seq). Ethylene emission, flesh firmness, or crispness of fruit from 1,273 F1 plants M. asiatica Nakai ‘Zisai Pearl’ × ‘Golden Delicious’ phenotyped prior to during 6 wk room-temperature storage. Segregation ethylene emission firmness traits was detected population. Thirteen QTL, including three major ones, on chromosome 03, 08, 16. A candidate gene encoding pectin acetylesterase, MdPAE10, QTL Z16.1 negatively affected life. 379-bp deletion coding sequence MdPAE10 disrupted its function. single nucleotide polymorphism (SNP) promoter region reduced transcription activity. These findings provided insight into control can be potentially used marker-assisted selection.

Null mutation of the MdACS3 gene, coding for a ripening-specific 1-aminocyclopropane-1-carboxylate synthase, leads to long shelf life in apple fruit.
Expression of MdACS1, coding for 1-aminocyclopropane-1-carboxylate synthase (ACS), parallels the level of ethylene production in ripening apple (Malus domestica) fruit. Here we show that expression of another ripening-specific ACS gene (MdACS3) precedes the initiation of MdACS1 expression by approximately 3 weeks; MdACS3 expression then gradually decreases as MdACS1 expression increases. متن کاملAn Antisense Pectin Methylesterase Gene Alters Pectin Chemistry and Soluble Solids in Tomato Fruit.
Pectin methylesterase (PME, EC 3.1.11) demethoxylates pectins and is believed to be involved in degradation of pectic cell wall components by polygalacturonase in ripening tomato fruit. We have introduced antisense and sense chimeric PME genes into tomato to elucidate the role of PME in fruit development and ripening. Fruits from transgenic plants expressing high levels of antisense PME RNA sho...
متن کاملEffects of Apple Juice Concentrate, Blackcurrant Concentrate and Pectin Levels on Selected Qualities of Apple-Blackcurrant Fruit Leather
A study was conducted to determine the effects of different levels of apple juice concentrate (AJC), blackcurrant concentrate (BCC) and pectin on the moisture content, water activity, color, texture and ascorbic acid content of apple-blackcurrant fruit leather using the response surface methodology. The results showed the moisture content increased with increasing pectin level and with greater ...
متن کاملApple Fruit Pectic Substances.
1. The pectic substances of apple have been extracted and separated into a pure pectinic acid and a neutral arabinan-galactan complex by precipitation of the acidic component with ethanol and with cetylpyridinium chloride. 2. The composition of the fractions has been determined. The pectinic acid contained galacturonic acid, arabinose, galactose, rhamnose, xylose and several trace sugars. 3. Tr...
